Micropropagation of Gypsophila paniculata through Shoot Tip Culture in Vitro



Abstract
In order to establish mass propagation system of Gypsophila paniculata by in vitro shoot tip culture, effects of growth regulators and ratio of NO©ý-N/NH©þ-N on the multiplication and growth of shoot were evaluated. Among cytokinins, BA was the most effective in shoot multiplication at 0.5¡­2.0§·/§¤ concentration, however vitrified plantlets increased compared with kinetin or 2iP treatment. Combination of BA and IAA ranging of 0.1¡­0.3§·/§¤ was better in shoot growth than BA treatment alone, bnt addition of growth regulators to media increased the vitrification of plantlets than control. Increasing ratio of NO©ý-N/NH©þ reduced number of vitrified plantlets and fresh/dry weight, while low NO©ý-N/NH©þ-N increased vitrified ones. Addition of 1.0§·/§¤ IBA into the media was effective in root formation of regenerated shoot in vitro. Survival rate of more than 93% of regenerated plants transferred in vivo condition, were resulted in perlite or mixture of perlite and vermiculite(1 : 1).
